Since gas is collected in a graduated tube above the open Mercury manometer so the pressure of the gas is  external pressure minus height of the Mercury in the tube
Pressure of the gas is equal to (750 - 100) mm Hg
Using combined gas law we can work out the volume of gas at STP
P1 V1/ T1 = P2V2/T2
